12 Hallmarks of Aging

Disabled macroautophagy

Hallmark 5 of 12 · Primary tier

Cellular self-cleaning slows, so damaged components and organelles are not cleared efficiently.

Autophagy clears protein aggregates, lipid droplets, and defective mitochondria via mitophagy. Declining autophagic flux with age links to sarcopenia, neurodegeneration, and immune senescence; chronic mTORC1 activity suppresses autophagy initiation.

How to minimize disabled macroautophagy

  • Periodic fasting, fasting-mimicking diets, or protein cycling can restore autophagy markers in human studies—effects are indirect but reproducible.
  • Exercise robustly induces autophagy in muscle and brain in animal models; human data are supportive though less direct.
  • Avoid chronic overnutrition and late-night eating, which sustain mTORC1 signaling and suppress autophagy.
  • Compounds like spermidine and rapamycin analogs induce autophagy preclinically; human longevity outcomes remain unproven.
