Loss of proteostasis
The cell's ability to fold, repair, and recycle proteins declines, allowing damaged proteins to build up.
Misfolded protein accumulation overwhelms chaperones and the ubiquitin–proteasome system, contributing to cataracts, sarcopenia, and neurodegeneration. Proteostasis collapse is a shared feature of Alzheimer's, Parkinson's, and Huntington's pathology.
How to minimize loss of proteostasis
- Induce heat-shock response: regular exercise and heat exposure (where medically safe) upregulate HSP70/90.
- Support the autophagy–proteostasis axis with adequate sleep, fasting windows, and balanced protein intake—not chronic excess.
- Reduce proteotoxic load: metabolic syndrome, alcohol, and chronic ER stress worsen misfolding burden.
- Pharmacologic chaperones remain mostly disease-specific and investigational for healthy aging.
